We unexpectedly found a structural resemblance between 7TMICs and the PHTF protein family, a deeply conserved group of proteins with unknown function, whose human homologs show elevated expression in the testis, cerebellum, and muscle. Our investigation also reveals divergent clusters of 7TMICs in insects, categorized as gustatory receptor-like (Grl) proteins. Subset-specific expression of Grls within taste neurons of Drosophila melanogaster suggests a previously unknown function as insect chemoreceptors. Our research, while acknowledging the possibility of convergent structural evolution, indicates a common eukaryotic origin for 7TMICs, countering the previously held belief of complete loss in Chordates, and illustrating the high adaptability of this protein fold, which likely explains its diverse functionalities in distinct cellular environments.
